ABSTRACT

To improve and broaden the antimicrobial activity of β-defensin130, 3 copies of β-defensin130 encoding sequences were synthesized and cloned into pET28a (+) expression vector, and expressed in Escherichia coli BL21 (DE3) as a 25 kDa soluble protein. The affinity purified 3×β-defensin 130 displayed antimicrobial activity against not only Gram-positive strains including Staphylococcus aureus (ATCC 25923) (45 μg/mL) and Listeria monocytogenes (ATCC 221633) (80 μg/mL) but also Gram-negative strains. Furthermore, the antimicrobial activity of β-defensin130 was not affected by temperature, pH and proteinase digestion. In addition, E. coli-derived 3×β-defensin130 was not toxic to HEK 293 cells and showed a relatively low hemolytic activity against rabbit erythrocytes. Our study proves 3×β-defensin130 expressed in E. coli is stable, non-cytotoxic and low-hemolytic active with great potential as alternative antibiotics.

ABSTRACT

Objective To investigate the effect of irrigation with hypothermic artificial cerebral-spinal fluid (aCSF)on expression of aquaporin-4(AQP-4) in the spinal cord following spinal ischemia-reperfusion (I/R) in rabbits.Methods Fifty-four adult male New Zealand white rabbits weighing 2.0-2.5 kg were randomly divided into 3 groups (n=18 each):group sham operation(S group); group I/R and group irrigation with hypothermic aCSF (FI group). Spinal I/R was induced by clamping the abdominal aorta below renal artery for 60 min. Hypothermic aCSF(25 X.)was infused at L4,5 interspace at a rate of 30 ml/h and drained from L7,8 interspace during spinal ischemia.Neurological function was evaluated at 4, 24, 48 and 72 h of reperfusion and scored (0=no hind limb activity, 4=hind limb function completely recovered) in 6 animals in each group. Six animals were sacrificed at 4, 24 and 72 h respectively in each group.The lumbar segment (L5-8) was removed for measurement of water content and AQP-4 protein expression (by immuno-histochemistry).Results Neurological function scores were significantly lower,water content was higher and AQP-4 expression smaller in group I/R than in group S. I/R-induced effects were significantly attenuated by irrigation of hypothermic aCSF. Conclusion Irrigation with hypothermic aCSF can ameliorate the spinal cord I/R injuries by up-regulation of AQP-4 expression.

ABSTRACT

Objective To observe the clinical effects and intubation condition of different doses of domestic cisatricurium(CA) in children. Methods Forty-five peadiatric patients aged 2 to 12 years old scheduled for selective surgery were randomized to three groups of 2 ED_(95) CA(0. 08 mg/kg), 3ED_(95) CA(0. 12 mg/kg) and 4ED_(95) CA(0. 16 mg/kg) (C_2, C_3, C_4 groups). Neuromuscular blockade was monitored with TOF-Guard. The conditions for trachea[intuDation were scored and the onset time, clinical duration, recovery index were recored. ResultsTracheal intubation was performed successfully in all patients. The conditions for tracheal intubation were scored greater 5 points in groups of C_3 and C_4 ,which was only 33. 3% of patients in group C_2. The onset time was shortened and the duration of action was prolonged as the dose of CA increased. But the recovery index of three groups were smilar. Conclusion An increase of CA dosage can shorten the onset time and prolong the duration of action and 3ED_(95) CA is suitable for tracheal intubation in children.

ABSTRACT

objective To analyze the causes of defective nonunion of femoral shaft and to evaluate interlocking intramedullary nails in treatment of shortened limb deformity following nonunion of femoral shaft. Methods 12 patients with shortened limb deformity following nonunion of femoral shaft were treated with autograft of ilium to fill up the defects and fixation by interlocking intramedullary nails. The patients were followed up for an average of 20 months. Results All the 12 patients healed by first intention after an average of 22 months. The limbs were lengthened averagely by 3.2cm. No failure occurred due to bent or broken major nails or intramedullary nails. Conclusions The main cause of shortened limb deformity following femoral shaft nonunion is bone resorption due to unreliable fixation. Interlocking intramedullary nails are an ideal device for shortened limb deformity following femoral shaft nonunion, though the union process is slow. In bone graft, care should be taken to avoid the complication of bone defect and weight bearing should be carried out some time later.
